Resuspension Calculator
The Resuspension Calculator determines how much diluent (buffer or nuclease-free water) you need to dissolve oligonucleotides to a target concentration. It is widely used in molecular biology for primer resuspension, oligo stock preparation, and PCR setup.
The resuspension formula relates oligo amount in nanomoles, desired concentration in micromolar, and diluent volume in microliters:
$$V_{\mu L} = \frac{n_{\mathrm{nmol}} \times 1000}{C_{\mu M}}$$
For example, to resuspend 60 nmol of oligo at 40 μM, you need $(60 \times 1000) / 40 = 1500$ μL of diluent. After adding the oligo, pipette or vortex to ensure complete dissolution.

Frequently Asked Questions
What is oligo resuspension?
Oligo resuspension is the process of dissolving lyophilized oligonucleotides in a buffer or nuclease-free water to create a stock solution at a known concentration for PCR, sequencing, or gene expression work.
What buffer should I use for resuspension?
TE buffer (Tris-EDTA) or nuclease-free water are common choices. TE protects DNA from degradation; water is preferred when EDTA would interfere with downstream enzymatic reactions.
How do I calculate diluent volume from nmol and μM?
Multiply the oligo amount in nmol by 1000, then divide by the desired concentration in μM. The result is the diluent volume in μL.
What concentration should I resuspend primers at?
Common stock concentrations are 100 μM for long-term storage or 10-50 μM for working stocks. Choose based on your PCR protocol and storage needs.
